OBJECTIVE:
1.1
To lay down a procedure for operation of Manifold Filtration Apparatus.
2.0
SCOPE:
2.1
This procedure is applicable to Manifold Filtration Apparatus located in
microbiology laboratory at Pharma Private Limited.

Microbiologist: Responsible for operation of the instrument.

Section In-charge: Responsible for review the
procedure

QC Head: Responsible for implementing the Procedure.
3.4
QA Head: Overall compliance.
4.0
PROCEDURE:
4.1
Ensure that the instrument is clean before starting.
4.2
Connect the manifold
filtration flushing tube end to the suction flask with hose pipe.
4.3
Connect the other end of the suction flask with vacuum
pump through hose pipe.
4.4
Place the sterile membrane filters on the sterile
filters holder and fix to the filtration Funnel.
4.5
Arrange the filtration funnel to the manifold
filtration flushing tube.
4.6
Pour required quantity of the sample in the filtration funnel and start the
vacuum pump, filtration will start and filtrate will collect in the suction
flask.
4.7
After completion of filtration remove the filter from the filter holder and
transfer to the sterile agar media plates.
4.8
Take another sterile filtration funnel to filter the
next sample.
4.9
After completion of filtration of all samples switch
off the vacuum pump mains and remove
the filtration funnels from flushing tube.
4.10
Cleaning:
4.10.1
Clean all parts with hot water by using smooth brush.
4.10.2
After cleaning rinse all the parts with Milli-Q water
or purified water.
4.11
Precautions:
4.11.1
Do not use steel wool or abrasive materials for cleaning since it will
causes scratches over the components.
4.11.2
Do not wipe components with dry paper or
cloth since fibres and lint left on Surfaces can generate electrostatic forces
that attract more dirt.
4.12
Before starting the filtration activity sanitize the
manifold filtration flushing tube with 70% IPA and sterilize the filtration
funnels and suction flask at 121°C for 15 minutes in an autoclave.
