1.0       OBJECTIVE:

To provide guidelines for Operation & Maintenance of G C System.

2.0       POLICY:  

To lay down a procedure of Operation and maintenance of G.C (Shimadzu 14 B)

3.0       PROCEDURE:

3.1       Switch on the Power supply and switch on stabilizer.

3.2       Switch on the instrument, heater, press FID button and Integrator, allow to warm the system for few minutes.           

3.3       Open the gas cylinders Helium, Air and Hydrogen by turning the opener key to the left slowly and gently.

3.4       Open the Toggle switch by lifting up Helium gas.

3.5       Feed the required temperatures to the instrument as per the method requirement Column Initial Temp, Injector and Detector Temperatures. Start the instrument

3.6                   Once the temperatures are attained heating indicators will blink and Ready Lamp will glow.

3.7                   Open the Air and Hydrogen toggle switches by lifting them. Adjust the pressure of Air and Hydrogen 50:60 ratio. Press the knob near the Air pressure dial and start igniting the detector with Continuous sparks gas lighter.  Check that the ignition has taken place by placing metallic piece just above the FID and see condensation.

3.8       Feed the required parameters to the integrator by pressing PARA and Enter.

3.9        Check for baseline. If the baseline is ok start analysis by injecting required Quantity with microlitre      
       syringe.
     
3.10     If the RSD of the three Standards chromatograms ratio is less than 2%, then the system is suitable for Sample injections. Otherwise repeat the Standard injections till you get the RSD below 2%.  

3.11     Inject the required Sample and take the chromatograms and calculate the Percentage of Active ingredient on the basis of respective peak ratios.

3.12     After completion of work, cool the instrument by calling cooling method File 4 and enter. Once the instrument is cool stop the gases and shutdown the power.

3.13     For better performance of GC system adhere to the following guidelines:

3.13.1  Use only required temperature as per the method.

3.13.2  Cool the instrument before shutting the power.

3.13.3  Close the gases, Check that all dials on the instrument come to Zero.

3.13.4    Do not open the chamber door during the working of the instrument.

3.14     Handle the injection syringe carefully. Do not pull the plunger out. Preserve carefully after use.

4.0       FREQUENCY:  As and when required.

5.0       RESPONSIBILITY:  Executive Q. A.

6.0       REVIEW GUIDE: Log register.
