1.0 OBJECTIVE:
To provide guidelines for Calibration of UV-Visible
Spectrophotometer
2.0 SCOPE
:
Periodically Calibration of UV- visible recording spectrophotometer
3.0 PROCEDURE
3.1
Switch on the instrument leave the
equipment to warm up for 15 min.
3.2 Dry Potassium Dichromate (A R grade) by
heating to constant weight at 1300C up to 3hrs.
3.3 Weigh
accurately 60.0 ± 3mg {NLT 57.0 mg & NMT 63.0 mg} and dissolve in
sufficient volume of 0.01N Sulfuric acid to produce 1000 ml.
3.4 Fill
the sampling cell with Potassium Dichromate solution, 0.01N Sulfuric acid is to
be filled in reference cell.
3.5 Control of Wave length: Verify the wavelength scale using the absorption
maxima of holmium perchlorate solution the line of hydrogen or deuterium
discharge lamp or the lines of a mercury vapour are shown below. The permitted
tolerance is ± 1 nm for the range 200 nm to 400nm and ± 3 nm for the range
400nm to 600nm.

3.6 Control
of absorbance: Measure the Absorbance of the Potassium Dichromate solution
at the following Wavelength, and calculate E. The extinction values (E) should
be within the limits specified in the Indian Pharmacopoeia.

3.7 Limit of stray light: Stray light may be detected at a given
wavelength with suitable filters or solution; for example, absorbance of a 1.2
per cent w/v solution of potassium chloride in a 1 cm cell should be greater
than 2.0 at about 200 nm when compared with water as reference liquid.
3.8 Resolution power: When stated in a monograph, record the spectrum of 0.02
per cent v/v solution of toluene in hexane UV. The ratio of the absorbance at
the maximum at about 269 nm to that at the minimum at about 266 nm is not less
than 1.5 unless otherwise specified in the monograph.
3.9 Cells: The absorbance of the cells intended to contain the
solution under examination and the reference liquid, when filled with the same
solvent should be identical. The tolerance on the path length of the cells used
is ± 0.005 cm cells should be cleaned and handled with great care.

Apart from extinction wavelength
calibration is done once yearly by the Shimadzu service
engineer.
3.11 The
instrument is covered under annual service contract and it is serviced as and
when there is a breakdown or requirement.

Internal Calibration once in three months for Control
of absorbance, Limit of stray light & Cells. Once in year for Control of
Wave length, Control of absorbance, Limit of stray light, Resolution power &
Cells by Equipment Service Engineer.
